Autonomous fueled mechanical replication of nucleic acid templates for the amplified optical detection of DNA

Yossi Weizmann, Zoya Cheglakov, Valeri Pavlov, Itamar Willner

(Figure Presented) Fuel for a DNA machine: A Fokl enzyme/DNA scission machine is triggered by the hybridization of its DNA hairpin structure with a target nucleic acid. The machine is fueled by a secondary fluorophore (F)- and quencher (Q)-functionalized nucleic acid 2. Scission of 3 leads to replication of the machinery cutter 1 and yields a fluorescent product 6, which provides an optical output for the machine (see scheme).
